Once you've pressed on Save icon, it gets gray, which means current note content was saved into the database.

By default it's every 3 minutes, and, on switching notes and on closing the app. Basically there is no way to avoid automatic Save command. This Save button is needed only to avoid data loss as result of sudden app or system crash, which is extremely rare thing.
If you want to configure auto-save, use menu item Tools -> Options, and in appeared window select Tools -> Backup, there you'll see "Auto-save every X minutes" option, so you can change the interval.

This is totally useless move, use File->Save As only when you want to change file location or its name, there is no other reason to use this command.
Basically with AllMyNotes you don't have to care about saving your data, the app will do everything for you, you are simply taking it too frequently, no need to worry about this topic at all. You can even hide this Save toolbar button as it has very little sense.
